چکیده

Sources of Compounds Tested-Thiamine hydrochloride (Merck, U.S.P. grade), 0-acetylthiamine hydrochloride and oxythiamine dihydrochloride (California Foundation for Biochemical Research), pyrithiamine hydrobromide and 4-amino-5 aminomethyl-a-methyl pyrimidine dihydrochloride (Nutritional Biochemicals Corporation) were all used without further purification. For determination of pK, values, the 4-amino-5-aminomethyl-2-methyl pyrimidine dihydrochloride was recrystallized from ethanol and ether until it gave the theoretical end point when titrated with base. 5-(2-Hydroxyethyl)I-methyl thiazole was redistilled from pooled commercial products of Merck’ and du Pont. The boiling point was lo%‘, 0.8 mm. Infrared inspection of the redistilled product showed the loss of a strong carbonyl peak which was originally present. The purified product was stored in a desiccator at -20” to hinder oxidation. Salts of 5-(d-Hydroxyethyl)-4-methyl Thiazole-The method of Clarke (6) was employed to prepare the 3-methyl and 3-benzyl (7) and the 3-o-, m-, and p-nitrobenzyl (8) salts. The 3-cr-methylbenzyl salt was prepared by dissolving 0.04 mole each of oc-bromoethyl benzene (Eastman, redistilled 89”, 15 mm.) and 5-(2-hydroxyethyl)-4-methyl thiazole in 4 ml. of thiophene-free anhydrous benzene in a stoppered 18 x 150-mm. test tube. The solution was heated overnight at 55”. The amethylbenzyl salt separates as a light yellow syrup. Attempts to crystallize this compound from a number of solvent systems were all unsuccessful. During crystallization attempts in hot absolute ethanol the salt was partially solvolyzed to the pyrimidylmethyl ethyl ether and 5-(2-hydroxyethyl)-4-methyl thiazole hydrobromide. The absorption spectra in both acid and base and the titration behavior very closely paralleled those of the 3-benzyl salt. These properties plus the method of preparation leave little doubt as to the character of the product. The 3-cyanomethyl salt was prepared in a similar manner from chloroacetonitrile (Eastman white label, redistilled 24O, 27 mm.) and 5-(2-hydroxyethyl)-4-methyl thiazole. The solution was heated at 80’ for 3 days, the benzene supernatant was discarded,

Pyruvate:ferredoxin oxidoreductase and the pyruvate dehydrogenase multi-enzyme complex both catalyse the CoA-dependent oxidative decarboxylation of pyruvate but differ in size, subunit composition and mechanism. Comparison of the pyruvate:ferredoxin oxidoreductase from the protozoon Trichomonas vaginalis and the pyruvate dehydrogenase component of the Escherichia coli pyruvate dehydrogenase com...

Effect of thiamine deficiency, pyrithiamine and oxythiamine on pyruvate metabolism in rat liver and brain in vivo.

Rats were fed either a thiamine-deficient diet of diets containing pyrithiamine or oxythiamine. When symptoms of thiamine deficiency appeared, the animals were injected intraperitoneally with [2-14C] pyruvate six to twelve minutes prior to sacrifice. Free glutamic and aspartic acids were isolated from liver and brain and degraded. The results indicate that, in thiamine-deficient or oxythiamine-...

PYRUVATE AND a-KETOGLUTARATE METABOLISM IN THIAMINE DEFICIENCY*

One obvious explanation for the effect of high fat diets in lowering the requirement for thiamine has been that fat metabolism does not require thiamine, while carbohydrate metabolism does. This explanation finds considerable support in one of the known functions of thiamine pyrophosphate. According to current theory, carbohydrate is oxidized to pyruvate, and pyruvate must be decarboxylated for...

Studies on Thiamine Analocues

Several analogues of thiamine’ are known to compete with the vitamin when administered to animals. Thus, Woolley and White (1) have observed that pyrithiamine caused the manifestation of typical thiamine deficiency symptoms, which could be competitively overcome by the vitamin. The antivitamin effect of oxythiamine in mice has been reported by Soodak and Cerecedo (2). Cerecedo et al. (3, 4), co...

Thiamine pyrophosphokinase deficiency in encephalopathic children with defects in the pyruvate oxidation pathway.

Thiamine pyrophosphate (TPP) is an essential cofactor of the cytosolic transketolase and of three mitochondrial enzymes involved in the oxidative decarboxylation of either pyruvate, α-ketoglutarate or branched chain amino acids. Thiamine is taken up by specific transporters into the cell and converted to the active TPP by thiamine pyrophosphokinase (TPK) in the cytosol from where it can be tran...

Disturbances of acetyl-CoA, energy and acetylcholine metabolism in some encephalopathies.

Acetyl-CoA provision to the synaptoplasmic compartment of cholinergic nerve terminals plays a regulatory role in the synthesis of acetylcholine. The disturbances in glucose utilization and in decarboxylation of the end product of its metabolism pyruvate, are considered to be significant factors causing cholinergic deficits in several diseases of the central nervous system. In this article we re...
